Lysosomes and Membrane Recycling

چکیده

A mechanism for intralysosomal membrane recycling is proposed. After invagination of the lysosomal membrane during autophagy, intralysosomal vesicles are formed. It is suggested that membrane can bleb out from these internal vesicles, probably in the form of very small vesicles, and return to the external lysosomal membrane by membrane fusion. This mechanism would conserve lysosomal membrane during autophagy, and is analogous to current models of plasma-membrane recycling. Its relationship to turnover of lysosomal-membrane proteins and other proteins is discussed.

Analyses of the recycling receptor, FcRn, in live cells reveal novel pathways for lysosomal delivery.

Lysosomes play a central role in the degradation of proteins and other macromolecules. The mechanisms by which receptors are transferred to lysosomes for constitutive degradation are poorly understood. We have analyzed the processes that lead to the lysosomal delivery of the Fc receptor, FcRn. These studies provide support for a novel pathway for receptor delivery. Specifically, unlike other re...

Diverse Autophagosome Membrane Sources Coalesce in Recycling Endosomes

Autophagic protein degradation is mediated by autophagosomes that fuse with lysosomes, where their contents are degraded. The membrane origins of autophagosomes may involve multiple sources. However, it is unclear if and where distinct membrane sources fuse during autophagosome biogenesis. Vesicles containing mATG9, the only transmembrane autophagy protein, are seen in many sites, and fusions w...

MARCH ubiquitin ligases alter the itinerary of clathrin-independent cargo from recycling to degradation

Following endocytosis, internalized plasma membrane proteins can be recycled back to the cell surface or trafficked to late endosomes/lysosomes for degradation. Here we report on the trafficking of multiple proteins that enter cells by clathrin-independent endocytosis (CIE) and determine that a set of proteins (CD44, CD98, and CD147) found primarily in recycling tubules largely failed to reach ...

A novel endocytic recycling signal that distinguishes the membrane trafficking of naturally occurring opioid receptors.

delta and micro opioid receptors are homologous G protein-coupled receptors that are differentially sorted between divergent degradative and recycling membrane pathways following agonist-induced endocytosis. Whereas delta opioid receptors are selectively sorted to lysosomes, micro opioid receptors recycle rapidly to the plasma membrane by a process that has been proposed to occur via bulk membr...

Hydrolase secretion is a consequence of membrane recycling

Acanthamoeba releases lysosomal hydrolases continuously into the culture medium. This release is specific for lysosomal hydrolases, but not other cellular proteins, and is energy dependent. The secreted hydrolases can be separated into two groups on the basis of their secretion kinetics: one is secreted at approximately 15% of the cellular activity per hour and the other at approximately 5%. In...
